For Singed's runes, the most important choice is our Quintessences. We want to use all three slots for Movement Speed Quintessences so he can constantly run around the map, create pressure, and cause chaos in fights. Magic Penetration Marks are ideal for making his poison deal more damage, while Armor Seals and Magic Resist Glyphs provide the defenses needed to keep him tanky and allow him to stay in fights longer.
Masteries
Singed is often one of the most focused targets in lane and during team fights since he has to get up close and personal to be effective. You want to max out the Defense tree to gain extra resistances, health, and damage reduction, allowing him to stay in fights longer while spreading his poison.
Singed still deals a large amount of damage, so put the remaining points into the Offense tree for additional Ability Power and Magic Penetration, helping his abilities hit even harder.

How to Play
With Singed you want to focus on farming during the early levels and avoid taking unnecessary fights before you've picked up a few items. Keep Q active while clearing waves and force your opponent to choose between chasing you or missing CS. If they overextend, use your E to throw them into your minion wave before taking them through your poison.
Don't be afraid to push the lane if you know where the enemy jungler is. You can even proxy farm behind the enemy tower, forcing your lane opponent to choose between farming under tower or chasing you and missing CS. Once level 6 arrives, your R gives you enough stats to play much more aggressively, making you very difficult to bring down.
