For runes, you want to focus on mobility, so Movement Speed Quintessences are a great choice. They allow you to position yourself better, dodge skill shots, and roam around the map more effectively to create picks for your team.
Magic Penetration Marks help increase the damage of your spells, making your abilities hit harder throughout the game. Armor Seals and Magic Resist Glyphs provide the defensive stats needed to survive the early game and allow you to scale safely into the late game.

While playing Ryze, your main goal is to scale into the late game and eventually take over the game. Since Ryze is relatively weak during the early stages, maxing out the Defense tree helps him survive the laning phase by providing extra resistances, health, and damage reduction, allowing him to stay in lane longer and farm safely.
Put the remaining points into the Offense tree, as Ryze still benefits greatly from the Cooldown Reduction, Ability Power, and Magic Penetration, helping him deal even more damage.
Arcane Mastery (P) When Ryze casts a spell, all other spells have their cooldown reduced by 1 second
Overload (Q) Passive/Active, Passive: Gain cooldown reduction. Active: Deal a % of Ryze's maximum mana
Rune Prison (W) Root the enemy
Spell Flux (E) Unleashes an orb that bounces to enemies or Ryze up to 5 times

Ryze is a scaling mage who wants to farm safely during the early game. Use your Q and W for short trades while also focusing on last hitting. Avoid unnecessary fights before you've picked up some mana items, as your damage scales heavily with your build.
You don't have great mobility and if using E your lane can push, so make sure to always ward. When you get your first item and ultimate, making trades and wave clearing will be much stronger. Don't worry if you get behind, Ryze scales really well going into the late game.
